The gene encoding Bcl-3 was first identified as the gene altered in some cases of chronic B cell lymphocytic leukemia and was later found to belong to the I kappa B subfamily of inhibitors of NF kappa B transcription factors. Bcl-3 does not have DNA-binding activity, but it interacts with NF kappa B1 and NF kappa B2 which, when bound to DNA, repress gene transcription. Therefore, Bcl-3 may promote gene expression that is repressed by members of the NF kappa B family. Bcl-3 RNA is highly expressed in spleen and liver with lower levels of expression in heart, skeletal muscle, lung, kidney, and testes.

This gene is a proto-oncogene candidate. It is identified by its translocation into the immunoglobulin alpha-locus in some cases of B-cell leukemia. The protein encoded by this gene contains seven ankyrin repeats, which are most closely related to those found in I kappa B proteins. This protein functions as a transcriptional co-activator that activates through its association with NF-kappa B homodimers.
