Goat anti-humnn IgA is conjugated to ptOTOxidase by means of a method described by Wilson MB and Nakane PK. (Reference: Wilson MB and Nakane PK. In Immunofluorescence and Related Staining Techniques, Elsevier/North Holland Biomedical Press, Amsterdam, P215 (1978).). Preparation of Diluent Buffer: Use 0.01 M TBS or 0.01 M PBS to dilute. See "Recommended Dilutions" below for details. Preparation of 0.01 M TBS: Add 1.2g Tris, 8.5g NaCI; 450ul of purified acetic acid or 700ul of concentrated hydrochloric acid to 1000ml H20 and adjust pH to 7.2-7.6. Finally, adjust the total Volume to 1L. Preparation of 0.01 M PBS: Add 8.5g sodium chloride, 1.4g Na2HP04 and 0.2g NaH2P04 to 1000ml distilled water and adjust pH to 7.2-7.6. Finally, adjust the total volume to 1L.
