Principle of the assay: mouse Angiopoietin-2 ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for Angiopoietin-2 has been precoated onto 96-well plates. Standards(CHO, M1-F496) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Angiopoietin-2 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse Angiopoietin-2 amount of sample captured in plate. Background: Angiopoietin-2 is a protein that in humans is encoded by the ANGPTL2 gene. Angiopoietins are members of the vascular endothelial growth factor family and the only known growth factors largely specific for vascular endothelium. Angiopoietin-1, angiopoietin-2, and angiopoietin-4 participate in the formation of blood vessels. Human and mouse ANGPT2 are 85% identical in amino acid sequence and approximately 60% identical to their ANGPT1 homologs. Transgenic overexpression of ANGPT2 in mice disrupted blood vessel formation in the mouse embryo. By FISH and radiation hybrid analysis, the human ANGPT2 gene was mapped to 8p23, and in a region of syntenic homology on mouse chromosome 8, where the Angpt2 gene was mapped. ANGPT2 is a naturally occurring antagonist of angiopoietin-1 that competes for binding to the TIE2 receptor and blocks ANGPT1-induced TIE2 autophosphorylation during vasculogenesis.
