products description :
Principle of the assay: human PAI-1 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for PAI-1 has been precoated onto 96-well plates. Standards(sf21, S22-P402) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for PAI-1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human PAI-1 amount of sample captured in plate. Background: Plasminogen activator inhibitor-1 is the principal inhibitor of tissue plasminogen activator (tPA) and urokinase (uPA), the activators of plasminogen and hence fibrinolysis (the physiological breakdown of blood clots). It is a serine protease inhibitor (serpin) protein (SERPINE1). PAI-1 is mainly produced by the endothelium (cells lining blood vessels), but is also secreted by other tissue types, such as adipose tissue. The PAI-1 gene is located on the seventh chromosome (7q21.3-q22).1 Congenital deficiency of PAI-1 leads to a hemorrhagic diathesis (a tendency tohemorrhage). PAI-1 is present in increased levels in various disease states (such as a number of forms of cancer), as well as in obesity and the metabolic syndrome. It has been linked to the increased occurrence of thrombosis in patients with these conditions. Additionally, it has been shown to interact with ORM1.2 The standard product used inthis kit is recombinant human PAI-1 with the molecular mass of 43KDa.

ncbi summary :
This gene encodes a member of the serine proteinase inhibitor (serpin) superfamily. This member is the principal inhibitor of tissue plasminogen activator (tPA) and urokinase (uPA), and hence is an inhibitor of fibrinolysis. Defects in this gene are the cause of plasminogen activator inhibitor-1 deficiency (PAI-1 deficiency), and high concentrations of the gene product are associated with thrombophilia. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2009]
uniprot summary :
SERPINE1: a secreted protein that acts as bait for tissue plasminogen activator, urokinase, and protein C. Its rapid interaction with TPA may function as a major control point in the regulation of fibrinolysis. Belongs to the serpin family. Interacts with VTN. Binds LRP1B; binding is followed by internalization and degradation. Plasma levels of PAI-1 and VCAM-1 together may be useful in predicting post-operative recurrence in patients with colorectal cancer. Protein type: Secreted; Motility/polarity/chemotaxis; Secreted, signal peptide. Chromosomal Location of Human Ortholog: 7q22.1. Cellular Component: extracellular matrix; extracellular space; plasma membrane; extracellular region. Molecular Function: serine-type endopeptidase inhibitor activity; protein binding; protease binding; receptor binding. Biological Process: circadian rhythm; extracellular matrix organization and biogenesis; positive regulation of blood coagulation; negative regulation of smooth muscle cell migration; defense response to Gram-negative bacterium; positive regulation of receptor-mediated endocytosis; positive regulation of interleukin-8 production; negative regulation of fibrinolysis; platelet degranulation; transforming growth factor beta receptor signaling pathway; regulation of receptor activity; angiogenesis; chronological cell aging; negative regulation of cell migration; negative regulation of cell adhesion mediated by integrin; transcription initiation from RNA polymerase II promoter; platelet activation; transcription, DNA-dependent; negative regulation of blood coagulation; regulation of cell proliferation; fibrinolysis; positive regulation of angiogenesis; gene expression; positive regulation of transcription from RNA polymerase II promoter; blood coagulation; positive regulation of inflammatory response.
