Intended Uses: This sandwich kit is for the accurate quantitative detection of human Interleukin 18 (also known as IL-18) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates. Principle of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human IL-18 antibody. IL-18 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human IL-18 Antibody is added and binds to IL-18 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated IL-18 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human IL-18. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL4) and interleukin 7 (IL7). The expression of this gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a similar gene in mice leads to ulcerative colitis-like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli.
