Intended Uses: This sandwich kit is for the accurate quantitative detection of human Cotinine (also known as Cotinine) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates. Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human Cotinine antibody. Cotinine present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human Cotinine Antibody is added and binds to Cotinine in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated Cotinine antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human Cotinine. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Tight junctions represent one mode of cell-to-cell adhesion in epithelial or endothelial cell sheets, forming continuous seals around cells and serving as a physical barrier to prevent solutes and water from passing freely through the paracellular space. These junctions are comprised of sets of continuous networking strands in the outwardly facing cytoplasmic leaflet, with complementary grooves in the inwardly facing extracytoplasmic leaflet. The protein encoded by this gene, a member of the claudin family, is an integral membrane protein and a component of tight junction strands. The encoded protein also binds specifically to the WW domain of Yes-associated protein. Defects in this gene are the cause of an autosomal recessive form of nonsyndromic sensorineural deafness. It is also reported that four synonymous variants in this gene are associated with kidney stones and reduced bone mineral density.
