products description :
TRPC6 Antibody: The mammalian transient receptor potential (TRP) superfamily can be divided into three major families including the "canonical TRP" (TRPC) family. The seven members of this family share the activation through PLC-coupled receptors and have been suggested to be components of receptor-regulated cation channels in different cell types. Furthermore, the members of the TRPC6/6/7 subfamily can be activated by diacylglycerol (DAG) analogs, suggesting a possible mechanism of activation of these channels by PLC-coupled receptors. When expressed in transfected cells, TRPC6 acts as a non-selective store-independent receptor-activated cation channel. It is activated by DAG in a PKC-independent manner and is insensitive to IP3 activation. There is increasing evidence that TRPC6 encodes endogenous DAG-activated receptor-operated cation channels in vivo.

ncbi summary :
The protein encoded by this gene forms a receptor-activated calcium channel in the cell membrane. The channel is activated by diacylglycerol and is thought to be under the control of a phosphatidylinositol second messenger system. Activation of this channel occurs independently of protein kinase C and is not triggered by low levels of intracellular calcium. Defects in this gene are a cause of focal segmental glomerulosclerosis 2 (FSGS2). [provided by RefSeq, Mar 2009]
uniprot summary :
TRPC6: Thought to form a receptor-activated non-selective calcium permeant cation channel. Probably is operated by a phosphatidylinositol second messenger system activated by receptor tyrosine kinases or G-protein coupled receptors. Activated by diacylglycerol (DAG) in a membrane-delimited fashion, independently of protein kinase C. Seems not to be activated by intracellular calcium store depletion. Interacts with MX1 and RNF24. Expressed primarily in placenta, lung, spleen, ovary and small intestine. Expressed in podocytes and is a component of the glomerular slit diaphragm. Belongs to the transient receptor (TC 1.A.4) family. STrpC subfamily. TRPC6 sub-subfamily. 3 isoforms of the human protein are produced by alternative splicing. Protein type: Membrane protein, integral; Channel, cation; Membrane protein, multi-pass. Chromosomal Location of Human Ortholog: 11q22.1. Cellular Component: plasma membrane; integral to membrane. Molecular Function: protein binding; store-operated calcium channel activity. Biological Process: positive regulation of ion transmembrane transporter activity; platelet activation; axon guidance; elevation of cytosolic calcium ion concentration; blood coagulation; positive regulation of calcium ion transport; transmembrane transport; cation transport; aging. Disease: Focal Segmental Glomerulosclerosis 2
