other info1 :
Conjugate: Unconjugated. Immunogen: NUMB antibody was raised against a 17 amino acid synthetic peptide near the carboxy terminus of human NUMB. Buffer: NUMB Antibody is supplied in PBS containing 0.02% sodium azide.
products description :
NUMB Antibody: NUMB, the mammalian homolog to the Drosophila asymmetric cell fate determinant NUMB, is thought to share several features molecular mechanisms in mammalian cells, generating asymmetric cell divisions during neurogenesis in vertebrate development as well as in hematopoietic stem cells. NUMB has been shown to inhibit Notch signaling, and is itself regulated by ubiquitinylation by MDM2. NUMB has also been shown to help activate the tumor suppressor p53, suggesting that loss of NUMB in cancerous cells would not only activate the potential oncogene Notch, but diminish the tumor suppressing effect of p53.

ncbi summary :
The protein encoded by this gene plays a role in the determination of cell fates during development. The encoded protein, whose degradation is induced in a proteasome-dependent manner by MDM2, is a membrane-bound protein that has been shown to associate with EPS15, LNX1, and NOTCH1. Four transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
uniprot summary :
NUMB: Plays a role in the process of neurogenesis. Required throughout embryonic neurogenesis to maintain neural progenitor cells, also called radial glial cells (RGCs), by allowing their daughter cells to choose progenitor over neuronal cell fate. Not required for the proliferation of neural progenitor cells before the onset of neurogenesis. Also involved postnatally in the subventricular zone (SVZ) neurogenesis by regulating SVZ neuroblasts survival and ependymal wall integrity. May also mediate local repair of brain ventricular wall damage. Interacts with CDH1 and TFAP2B. Interacts with EPS15, LNX and NOTCH1. May interact with DUOXA1. Interacts with RALBP1 in a complex also containing EPN1 and TFAP2A during interphase and mitosis. 4 isoforms of the human protein are produced by alternative splicing. Protein type: Cell development/differentiation. Chromosomal Location of Human Ortholog: 14q24.3. Cellular Component: extrinsic to plasma membrane; focal adhesion; clathrin-coated vesicle; apical part of cell; basolateral plasma membrane; integral to plasma membrane; early endosome; plasma membrane; nucleus. Molecular Function: protein binding; cadherin binding; beta-catenin binding; alpha-catenin binding. Biological Process: positive regulation of polarized epithelial cell differentiation; axon guidance; positive regulation of neurogenesis; Notch signaling pathway; negative regulation of Notch signaling pathway; neuroblast division in the subventricular zone; lateral ventricle development; positive regulation of cell migration
