Description: DUT Human Recombinant fused with a 21 amino acid His tag at N-terminus produced in E Coli is a single, non-glycosylated, polypeptide chain containing 204 amino acids (70-252 a.a.) and having a molecular mass of 21.6kDa. The DUT is purified by proprietary chromatographic techniques. Introduction: Deoxyuridine Triphosphatase (DUT) is a ubiquitous enzyme that functions in nucleotide metabolism. Deoxyuridine Triphosphatase, in the presence of magnesium ions, is responsible for hydrolyzing dUTP to dUMP and diphosphate. This reaction is imperative for keeping the intracellular dUTP concentration low so that uracil does not become incorporated into DNA. Extensive integration of uracil into DNA can eventually lead to cell death. This suggests that DUT is crucial for cell viability, further implying that it is a prospective target for anticancer therapy.

This gene encodes an essential enzyme of nucleotide metabolism. The encoded protein forms a ubiquitous, homotetrameric enzyme that hydrolyzes dUTP to dUMP and pyrophosphate. This reaction serves two cellular purposes: providing a precursor (dUMP) for the synthesis of thymine nucleotides needed for DNA replication, and limiting intracellular pools of dUTP. Elevated levels of dUTP lead to increased incorporation of uracil into DNA, which induces extensive excision repair mediated by uracil glycosylase. This repair process, resulting in the removal and reincorporation of dUTP, is self-defeating and leads to DNA fragmentation and cell death.
