products description :
Description: PDCD6IP Human Recombinant produced in E Coli is a single, non-glycosylated polypeptide chain containing 412 amino acids (1-392 a.a.) and having a molecular mass of 45.8 kDa. The PDCD6IP is fused to 20 amino acid His-tag at N-terminus and purified by conventional chromatography. Introduction: PDCD6IP is a Class E VPS protein which participates in concentration and sorting of cargo proteins of the multivesicular body or incorporation into intralumenal vesicles that are generated by invagination and scission from the limiting membrane of the endosome. PDCD6IP binds to the phospholipid lysobisphosphatidic acid which is abundant in MVBs internal membranes. The MVB pathway appears to require the sequential function of ESCRT-O, -I,-II and -III complexes. PDCD6IP is an adapter for a subset of ESCRT-III proteins, such as CHMP4, to function at distinct membranes. PDCD6IP is mandatory for completion of cytokinesis. PDCD6IP takes part in HIV-1 virus budding. PDCD6IP replaces TSG101 in its function of supporting HIV-1 release. PDCD6IP takes part in the regulation of both apoptosis and cell proliferation. PDCD6IP is a cytoplasmic protein that cooperates with apoptosis-associated proteins (ALG-2 and PDCD6) and with the endocytosis-regulator CIN85. Overexpression of PDCD6IP and endophilin

ncbi summary :
This gene encodes a protein that functions within the ESCRT pathway in the abscission stage of cytokinesis, in intralumenal endosomal vesicle formation, and in enveloped virus budding. Studies using mouse cells have shown that overexpression of this protein can block apoptosis. In addition, the product of this gene binds to the product of the PDCD6 gene, a protein required for apoptosis, in a calcium-dependent manner. This gene product also binds to endophilins, proteins that regulate membrane shape during endocytosis. Overexpression of this gene product and endophilins results in cytoplasmic vacuolization, which may be partly responsible for the protection against cell death. Several alternatively spliced transcript variants encoding different isoforms have been found for this gene. Related pseudogenes have been identified on chromosome 15. [provided by RefSeq, Jan 2012]
uniprot summary :
Alix: an adaptor protein that may regulate the function of receptor and cytoskeleton-associated tyrosine kinases. A class E vacuolar protein sorting (VPS) factor involved in concentration and sorting of cargo proteins of the multivesicular body (MVB) for incorporation into intralumenal vesicles. Fusion between endosomes and the vacuole will then target the cargo proteins to the vacuolar lumen. In case of infection, the HIV-1 virus directly takes advantage of its function for viral exocytosis and budding, via its interaction with HIV-1 p6 protein. May play a role in the regulation of both apoptosis and cell proliferation. Protein type: Apoptosis; Adaptor/scaffold. Chromosomal Location of Human Ortholog: 3p22.3. Cellular Component: focal adhesion; membrane; cytoplasm; melanosome; immunological synapse; microtubule organizing center; cytosol; vesicle. Molecular Function: protein binding; protein homodimerization activity; proteinase activated receptor binding; SH3 domain binding; calcium-dependent protein binding. Biological Process: protein transport; cell separation during cytokinesis; viral reproduction; apoptosis; viral infectious cycle; mitotic metaphase plate congression; nuclear organization and biogenesis
