products description :
Description: RHEB Human Recombinant produced in E Coli is a single, non-glycosylated polypeptide chain containing 197 amino acids (1-181 amino acids) and having a molecular mass of 21.7 kDa.The RHEB is fused to T7-tag at N-terminus (16 a.a.) and is purified by standard chromatography techniques. Introduction: RHEB is part of the Ras & GTPase superfamily that was originally identified as an immediate-early gene in brain but is also widely expressed in other tissues. RHEB encodes a lipid-anchored, cell membrane protein with five repeats of the RAS-related GTP-binding region. RHEB is necessary in regulation of growth and cell cycle progression due to its role in the insulin/TOR/S6K signaling pathway. RHEB has GTPase activity and shuttles between a GDP-bound form and a GTP-bound form, and farnesylation of the protein is required for this activity. RHEB induces oncogenic transformation. RHEB overexpression accelerates lymphomagenesis and is associated with prostate cancer. RHEB can cytopathologically distinguish between fibroadenoma from malignant breast carcinomas which is considered as a secondary diagnostic tool. RHEB has a central role in the regulation of the Ras/B-Raf/C-Raf/MEK signaling network.

ncbi summary :
This gene is a member of the small GTPase superfamily and encodes a lipid-anchored, cell membrane protein with five repeats of the RAS-related GTP-binding region. This protein is vital in regulation of growth and cell cycle progression due to its role in the insulin/TOR/S6K signaling pathway. The protein has GTPase activity and shuttles between a GDP-bound form and a GTP-bound form, and farnesylation of the protein is required for this activity. Three pseudogenes have been mapped, two on chromosome 10 and one on chromosome 22. [provided by RefSeq, Jul 2008]
uniprot summary :
RHEB: Stimulates the phosphorylation of S6K1 and EIF4EBP1 through activation of mTORC1 signaling. Activates the protein kinase activity of mTORC1. Has low intrinsic GTPase activity. Binds to mTORC1 in a guanyl nucleotide-independent manner. Interacts directly with MTOR, MLST8 and RPTOR. Interacts with TSC2. Ubiquitous. Highest levels observed in skeletal and cardiac muscle. Alternates between an inactive form bound to GDP and an active form bound to GTP. Inactivated by TSC1-TSC2 via the GTPase activating protein (GAP) domain of TSC2. Belongs to the small GTPase superfamily. Rheb family. Protein type: G protein; G protein, monomeric, Rheb; G protein, monomeric. Chromosomal Location of Human Ortholog: 7q36. Cellular Component: Golgi membrane; spliceosome; endoplasmic reticulum membrane; cell soma; membrane; dendrite; plasma membrane; cytosol. Molecular Function: GTPase activity; protein binding; GDP binding; GTP binding; metal ion binding; protein kinase binding. Biological Process: metabolic process; small GTPase mediated signal transduction; insulin receptor signaling pathway; cell cycle arrest; signal transduction; positive regulation of TOR signaling pathway
