Clone REA448 recognizes the human CD227 antigen, a single-pass type I membrane protein which is also known as mucin-1 (MUC-1), episialin, or epithelial membrane antigen (EMA). CD227 is a heterodimeric epithelial cell glycoprotein that is normally expressed at low levels on the apical surface of epithelial cells and on some hematopoietic cell lineages including subsets of T cells, B cells, monocytes, and dendritic cells. In most adenocarcinomas and in other inflammatory diseases of epithelial tissues, CD227 is overexpressed and loses the polarity of its expression. On normal epithelia, CD227 is extensively O-glycosylated with long branched glycans, whereas on tumor cells they are markedly hypoglycosylated with simpler and shorter glycan chains. Elevated levels of CD227 on the tumor have been associated with invasiveness and poor prognosis in colon, pancreas, breast, and bladder cancer. CD227 regulates the metabolite flux at multiple levels. Serving as a transcriptional co-activator, CD227 directly regulates expression of metabolic genes. By regulating receptor tyrosine kinase signaling, CD227 facilitates production of biosynthetic intermediates required for cell growth. CD227 also modulates the activity/stability of enzymes and transcription factors that directly regulate metabolic functions. Additional information: Clone REA448 displays negligible binding to Fc receptors.
