Endo S2, secreted by Streptococcus pyogenes, deglycosylates the conserved N-linked glycan at Asn297 on IgG Fc to eliminate its effector functions and evade the immune system. EndoS2 specifically hydrolyzes diverse glycans on the Fc region of IgG antibodies using a mechanism that relies on its glycoside hydrolase domain and a coevolved carbohydrate-binding module [1] [2] . Endo S2 Protein, S. pyogenes (His) is the recombinant Endo S2 protein, expressed by E. coli , with N-6*His labeled tag.

EndoS2 has a broader substrate glycan specificity, hydrolyzing not only biantennary CT N-glycans, but also high-mannose (HM), hybrid, and bisecting CT N-glycans on IgG [1] . Endo-β-N-acetylglucosaminidases (ENGases, EC 3.2.1.96) are a class of enzymes, which can hydrolytically cleave β-1,4 glycosidic bonds within the N,N′-diacetylchitobiose moiety in the inner-core region of N-glycan and release glycan chain from their associated proteins, thus leaving one unit of N-acetylglucosamine (GlcNAc), with or without the core fucose, linked to the asparagine residue [2] .
