Crlf1 protein exhibits the formation of covalent di- and tetramers, and it engages in the creation of a heteromeric complex with cardiotrophin-like cytokine CLCF1/CLC. This CRLF1-CLCF1 complex serves as a ligand for the ciliary neurotrophic factor receptor/CNTFR. Notably, the CRLF1-CLCF1 heterodimer, along with the tripartite signaling complex formed by CRLF1, CLCF1, and CNTFR, binds SORL1, where the interaction is predominantly mediated by the CRLF1 moiety within this complex. These intricate interactions underscore the multifaceted roles of Crlf1 protein in both neurodevelopmental processes and immune regulation.
