IGF1R is a receptor protein that plays a crucial role in cell growth and survival control. It is involved in tumor transformation and the survival of malignant cells. When a ligand, such as insulin-like growth factor 1 (IGF1), binds to IGF1R, it activates the receptor kinase, leading to autophosphorylation of the receptor and phosphorylation of multiple signaling adapter proteins, including insulin-receptor substrates (IRS1/2), Shc, and 14-3-3 proteins.Phosphorylation of IRS proteins activates two main signaling pathways: the PI3K-AKT/PKB pathway and the Ras-MAPK pathway. Activation of the MAPK pathway results in increased cellular proliferation, while activation of the PI3K pathway inhibits apoptosis and stimulates protein synthesis. Phosphorylated IRS1 can activate the regulatory subunit of PI3K (PIK3R1), which then activates downstream substrates, including protein AKT/PKB. AKT phosphorylation enhances protein synthesis through mTOR activation and triggers the antiapoptotic effects of IGF1R by phosphorylating and inactivating BAD.In addition to the PI3K-AKT/PKB and Ras-MAPK pathways, IGF1R also signals through the Janus kinase/signal transducer and activator of transcription pathway (JAK/STAT). Phosphorylation of JAK proteins leads to phosphorylation and activation of signal transducers and activators of transcription (STAT) proteins. The activation of STAT3, in particular, may be essential for the transforming activity of IGF1R. The JAK/STAT pathway activates gene transcription and may contribute to the transforming activity of IGF1R.The IGF1R can also activate JNK kinases. However, IGF1 exerts inhibitory effects on JNK activation by phosphorylating and inhibiting MAP3K5/ASK1, which can directly associate with IGF1R. Additionally, when IGF1R forms a hybrid receptor with the insulin receptor (INSR), it can bind IGF1.Overall, IGF1R plays a critical role in regulating cell growth, survival, and transformation through multiple signaling pathways, including the PI3K-AKT/PKB, Ras-MAPK, JAK/STAT, and JNK pathways.
