These heterodimers bind to the core sites of various enhancers and promoters, such as murine leukemia virus, polyomavirus enhancer, T-cell receptor enhancers, LCK, IL3, and GM-CSF promoters. CBF complexes play a role in the repression of the ZBTB7B transcription factor during cytotoxic (CD8+) T cell development, acting as a transcriptional silencer by binding to the RUNX-binding sequence within the ZBTB7B locus, thereby facilitating cytotoxic T cell differentiation. The interaction involves heterodimerization with RUNX1, RUNX2, and RUNX3, and CBFB also interacts with COPRS and forms a complex with PRMT5 and RUNX1.
