Neuroligin-3 (NLGN3) is a cell surface protein that plays a crucial role in cell-cell interactions through its interactions with neurexin family members. It is involved in synapse function and synaptic signal transmission, potentially by clustering other synaptic proteins. While NLGN3 may promote the initial formation of synapses, it is not essential for this process. Additionally, it may be involved in glia-glia or glia-neuron interactions in the developing peripheral nervous system. NLGN3 forms homodimers and heterodimers with NLGN1 and NLGN2, and interacts with neurexins NRXN1, NRXN2, and NRXN3, with the interaction being mediated by heparan sulfate glycan modification on neurexin. Furthermore, NLGN3 interacts with DLG4/PSD-95 through its C-terminus, specifically with the PDZ domain 3 of DLG4/PSD-95.
