CSEN is a calcium-dependent transcriptional repressor that binds to the DRE elements of genes such as PDYN and FOS. Its affinity to DNA is affected by calcium. Beyond its transcriptional functions, CSEN acts as a regulatory subunit of Kv4/D (Shal)-type voltage-gated rapidly inactivating A-type potassium channels, such as KCND2/Kv4.2 and KCND3/Kv4.3. In this capacity, it modulates channel expression at the cell membrane, influencing gating characteristics, inactivation kinetics, and the rate of recovery from inactivation. Notably, these regulatory actions are exerted in a calcium-dependent and isoform-specific manner, underscoring the complexity of CSEN's role in potassium channel activity.
