The B7-H4 protein acts as a negative regulator, inhibiting T cell activation, proliferation, cytokine production, and cytotoxicity. When expressed on tumor macrophages, it cooperates with regulatory T cells (Tregs) to suppress tumor-associated antigen-specific T cell immunity and create an immunosuppressive microenvironment.
