TEAD3, a transcription factor, assumes a pivotal role in the Hippo signaling pathway, a regulatory network crucial for organ size control and tumor suppression by orchestrating proliferation inhibition and apoptosis promotion. MOB1 subsequently phosphorylates and inactivates the YAP1 oncoprotein and WWTR1/TAZ. TEAD3 functions by mediating the gene expression of YAP1 and WWTR1/TAZ, thereby regulating cell proliferation, migration, and epithelial-mesenchymal transition (EMT) induction. Additionally, TEAD3 is involved in binding to multiple functional elements of the human chorionic somatomammotropin-B gene enhancer. Its interactions with YAP1 and WWTR1/TAZ underscore its central role in the regulatory dynamics of this signaling pathway.
