The S100A4 protein is a calcium-binding protein involved in angiogenesis, cell differentiation, apoptosis, and autophagy. S100A4 protein interacts with MYH9 to enhance cell motility and invasion. S100A4 also inhibits the pro-apoptotic function of TP53 by reducing its protein levels. Functionally, it enhances cell motility and invasiveness by interacting with non-muscle myosin heavy chain IIA/MYH9, promoting filament depolymerization and increasing the amount of soluble myosin-IIA to facilitate chemotaxis. Additionally, S100A4 modulates the pro-apoptotic function of TP53 by binding to its C-terminal transactivation domain within the nucleus, leading to a reduction in TP53 protein levels. In the extracellular space, it stimulates cytokine production and acts as a chemoattractant complex with PGLYRP1 to promote lymphocyte migration via CCR5 and CXCR3 receptors. The protein forms homodimers and interacts with various partners, including PPFIBP1, Annexin 2/ANXA2, TP53, CCR5, and FCGR3A, each interaction contributing to its diverse cellular functions.
