Vinculin has no enzymatic activity. It regulates adhesion by directly binding to actin, stimulating actin polymerization and recruiting actin remodeling proteins. In the absence of vinculin, cell-matrix and cell-cell adhesion are dramatically impaired, indicating vinculin plays a critical role in human physiology. Vinculin stabilizes focal adhesions and thereby suppresses cell migration, an effect that is relieved by transient changes in the local concentrations of inositol phospholipids. Vinculin provides a mechanical link, controls cell signalling processes, affects contractility and adhesion protein turnover. Vinculin is thought to function as a tumour suppressor by supporting anchorage-dependent cell growth and suppressing tumour metastasis by reducing cell motility. In addition, Vinculin regulates cell-surface E-cadherin expression and potentiates mechanosensing by the E-cadherin complex, may also play important roles in cell morphology and locomotion [1] [2] [3] [4] .
