Within PI3KC3-C1, PK3C3 actively promotes the formation of endoplasmic reticulum membrane curvature before vesicle budding. Furthermore, PK3C3 is implicated in the regulation of degradative endocytic trafficking, essential for the abscission step in cytokinesis, likely in the context of PI3KC3-C2. Additionally, PK3C3 plays a crucial role in the transport of lysosomal enzyme precursors to lysosomes and is required for the progression from early to late endosomes. Notably, the kinase activity of PK3C3 is vital for the replication of SARS coronavirus-2 (SARS-CoV-2) during microbial infection.
