IL-24 protein, produced by T-cells, regulates immune responses, tissue homeostasis, defense, and oncogenesis. It induces type I interferon response in influenza infection, signaling through IL20RA/IL20RB or IL20RB/IL22RA1 receptor complexes to stimulate JAK1-STAT3 and MAPK pathways. IL-24 promotes secretion of IL8 and MMP1, maintains ER homeostasis, and serves as a quality control mechanism for the ubiquitin proteasome system. IL-24 protein, a multifunctional cytokine primarily produced by T-cells, orchestrates a regulatory role in immune responses, tissue homeostasis, host defense, and oncogenesis. Demonstrating antiviral functions, IL-24 induces the type I interferon response during influenza infection. The cytokine signals through two receptor complexes, IL20RA/IL20RB or IL20RB/IL22RA1, stimulating the JAK1-STAT3 and MAPK pathways, thereby promoting the secretion of pro-inflammatory mediators, including IL8 and MMP1. Intracellularly, IL-24 maintains endoplasmic reticulum homeostasis by constraining the eIF2alpha-CHOP pathway-mediated stress signal. Additionally, it acts as a quality control mechanism for the ubiquitin proteasome system, detecting proteasome dysfunction and activating PKR/EIF2AK2.
