ELISA Capture: Purified monoclonal antibody is the preferred capture antibody in a sandwich ELISA for quantifying human and/or viral IL-10 proteins.1-5 Purified antibody should be paired with biotinylated monoclonal antibody as the detection antibody, with purified recombinant human IL-10 as the standard. Immunofluorescence/. Flow Cytometry: The antibody is useful for intracytoplasmic staining and flow cytometric analysis to identify and enumerate IL-10-positive cells within mixed cell populations. Recombinant human IL-10 or unlabeled Rat IgG1 should be used as a specificity control. Western Blot: The purified antibody is also useful for Western blotting applications. A concentration of 1-5 ug/ml (in conjunction with AP-labeled Goat Anti-Rat-Ig) has been found to enable detection of 100 ng/lane of recombinant human IL-10 under reducing conditions.
