Suitable for use in ELISA, Western Blot, Immunohistochemistry and Neutralization. Neutralization of Human IFN-g R1 Bioactivity: The exact concentration of antibody required to neutralize rhIFN-g R1-mediated bioactivity is dependent on the cytokine concentration, cell type, growth conditions and the type of activity studied. The Neutralization Dose50 (ND50) for this antibody is defined as that concentration of antibody required to yield one-half maximal inhibition of the cytokine activity on a responsive cell line, when that cytokine is present at a concentration just high enough to elicit a maximum response. The ND50 is ~0.5-2 ug/ml in the presence of 2 ng/ml of rhIFN-g, using a confluent culture of HeLa cells. ELISA (Direct): 0.5-1 ug/ml. The detection limit for rhIFN-g R1 is ~0.3 ng/well. Western Blot: 0.1-0.2 ug/ml. The detection limit for is ~5 ng/lane under non-reducing and reducing conditions. Immunohistochemistry: 15 ug/ml. Detects IFN-g R1 in human PBMC's (peripheral blood mononuclear cells) (fluorescence immunocytochemistry).
