UsageText :
Western Blot, Immunohistochemistry, Flow Cytometry, Blocking of receptor-ligand interaction, Direct ELISA. Western Blot: This antibody can be used at 0.1-0.2 ug/mL with the appropriate secondary reagents to detect mouse PD-L2. The detection limit for rmPD-L2 is approximately 2 ng/lane under non-reducing and reducing conditions. Immunohistochemistry: This antibody will detect PD-L2 in cells and tissues. The working dilution is 2-15 ug/mL. Flow Cytometry: This antibody has been tested on Tim-1 stimulated RAW 264.7 cells for use in flow cytometry. Dilute this antibody to 50 ug/mL and add 10 ul of the diluted solution to 1-2.5 x 10^5 cells in a total reaction volume not exceeding 200ul. The binding of unlabeled antibodies may be visualized by adding 10 ul of a 25 ug/mL stock solution of a secondary developing reagent such as anti-goat IgG conjugated to a fluorochrome. Blocking of receptor-ligand interaction: Approximately 1-4 ug/mL of this antibody will block 50% of the binding of 1 ug/mL of rmPD-1 to immobilized rmPD-L2/Fc chimera (100 ul of a 1 ug/mL solution was coated in each well) in a functional ELISA. Direct ELISA: This antibody can be used at 0.5-1.0 ug/mL with the appropriate secondary reagents to detect mouse PD-L2. The detection limit for rmPD-L2 is approximately 1.2 ng/well.
