Suitable for use in Immunocytochemistry, Western blot, and Direct ELISA. Western Blot: 1-2 ug/ml. Detection limit is ~1 ng/lane under non-reducing and. reducing conditions. Chemiluminescent detection with WesternGlo Chemiluminescent. Detection Substrate will increase sensitivity by 5 to 50 fold. Immunocytochemistry:-15 ug/ml will detect Mash1 on retinoic acid-differentiated human NTERA-2 cells. Cells were fixed with PBS containing 4% paraformaldehyde for 20 minutes at RT and blocked with PBS containing 10% normal donkey serum, 0.1% Triton X-100, and 1%. BSA for 45 minutes at RT. After blocking, cells were incubated with diluted primary antibody overnight at 4C followed by Rhodamine Red-coupled anti-rat IgG at RT in the dark for one hour. Wash cells with PBS containing 0.1% BSA. Direct ELISA: 0.5-1.0 ug/ml The detection limit 1 is 0.5 ng/well.
