Suitable for use in ELISA, Western Blot, Immunocytochemistry and Immunohistochemistry. Western Blot: 0.1-0.2 ug/ml. Detection limit for rhVersican is ~2 ng/lane under non-reducing and reducing conditions. Immunocytochemistry: 10 ug/ml detects Versican in Hela cells. Cells were fixed with PBS containing 4% paraformaldehyde for 20 minutes at RT and blocked with PBS containing 10% normal donkey serum, 0.1% Triton X-100, and 1% BSA for 45 minutes at RT. After blocking, cells were incubated with diluted primary antibody overnight at 4? C followed by fluorochrome-coupled anti-goat IgG at RT in the dark for one hour. Between each step, cells were washed with PBS containing 0.1% BSA. Immunohistochemistry: 10 ug/ml detects Versican in cryosections of E13.5 mouse embryos. Tissue was fixed with PBS containing 4%PFA for 20 min. at RT and blocked with PBS containing 10% normal donkey serum, 0.1% Triton and 1% BSA for 45 minutes at RT. After blocking, sections were incubated with diluted primary antibody for 3 hours at RT followed by fluorochrome-coupled anti-goat lgG at RT for 1 hour. Between each step, sections were washed with 0.1% BSA in PBS. Direct ELISA: 0.5-1.0 ug/ml. Detection limit for rhVersican is ~0.1 ng/well. Optimal dilutions should be determined by each laboratory.
