UsageText :
Suitable for use in Western Blot, Flow Cytometry, Immunohistochemistry and ELISA. Western Blot: 1-2 ug/ml. Detection limit is approximately 25 ng/lane under non-reducing conditions. Reducing conditions are not recommended. Flow Cytometry: 25 ug/ml. For intracellular staining, cells must be fixed and permeabilized using 4% paraformaldehyde and 0.1% saponin in PBS. Add 10 ul of the diluted solution to 1-5x10^5 cells in a total reaction volume not exceeding 200ul. Incubate for 30 minutes and then wash cells with 0.1% saponin prior to addition of a secondary developing reagent. Cells should be washed a final time in 0.1% saponin prior to flow cytometric analysis. Direct ELISA: 0.5-1 ug/ml. Detection limit is ~5 ng/well. Immunohistochemistry: 8-25 ug/ml with appropriate secondary reagents to detect IL-17D in paraffin-embedded normal heart tissue sections.
