Western Blot Analysis 2: At 0.5-2 ug/ml, this antibody detected Cyclin D in a RIPA lysates of human A431 cells, mouse 3T3 fibroblasts. Positive Antigen Control: EGF-stimulated A431 cell lysate. Use 20 ug per lane. Immunoprecipitation: 5 ug of antibody immunoprecipitated Cyclin D from a RIPA lysate of human A431 cells. An H1 kinase assay can be performed with the immunoprecipitate, as reported previously. Immunocytochemistry: At 10 ug/ml, a previous lot of antibody detected Cyclin D in cells fixed with 4% paraformaldehyde/0.1% Triton X-100. Western Blot Analysis EGF-stimulated A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-Cyclin D (2 ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.
