The 16a11 antibody has been tested by flow cytometric analysis of C57Bl/6 and BALB/c mouse splenocyte suspensions and can be used at less than or equal to 1 ug per test. A test is defined as the amount (ug) of antibody that will stain a cell sample in a final volume of 100 ul. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. For optimal detection of NKG2A^B6 using 16a11, a fluorochrome conjugated isotype-specific second step is recommended. If using a polyclonal anti-mouse IgG second step, please note that splenic B cells will also stain due to binding of the second step to surface IgG. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
