Immunohistochemistry: LS-B3028 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B3028 was determined to be 4 ug/ml. Peptide ELISA: antibody detection limit dilution 1:32000. Western Blot: Approx 35kD band observed in Human lysates of cell lines A431 and K562 (calculated MW of 38.7kD according to NP_000691.1). The observed molecular weight corresponds to earlier findings in literature with different antibodies (Paweletz et al, Cancer Res. 2000 Nov15;60(22):6293-7. PMID: 11103786). Recommended concentration: 0.5-1.5 ug/ml.
