Western blot of lysate from HT1080 cell line,using IFITM3 Antibody. Antibody was diluted at 1:1000 at each lane. A goat anti-rabbit IgG H&L (HRP) at 1:5000 dilution was used as the secondary antibody.Lysate at 35ug per lane.

Fluorescent confocal image of HeLa cells stained with IFITM3 antibody. HeLa cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min), then incubated with Fragilis (IFITM3) primary antibody (1:200, 2 h at room temperature). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (5.25 mu M, 25 min). Nuclei were counterstained with Hoechst 33342 (blue) (10 ug/ml, 3 min). Note the highly specific localization of the IFITM3 immunoreactivity to the Golgi.

Immunofluorescent analysis of IFITM3 staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
