Western blot analysis of LT-alpha (C-term) on different lysates with Rabbit anti-LT-alpha (C-term) antibody (0807-6) at 1/2,000 dilution. Lane 1: Ramos cell lysate Lane 2: Mouse skeletal muscle tissue lysate (negative) Lane 3: Mouse spleen tissue lysate Lane 4: Rat spleen tissue lysate Lysates/proteins at 20 µg/Lane. Predicted band size: 22 kDa Observed band size: 26 kDa Exposure time: 30 seconds; ECL: K1801; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (0807-6) at 1/2,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Standard curve of TNF-β matched pair antibodies: Sandwich ELISA analysis of TNF-β matched pair antibodies Elisa assay was performed by coating wells of a 96-well plate with 50 µl per well of capture antibody HA721242 [PS01-05] diluted in carbonate/bicarbonate buffer, at a concentration of 2 µg/mL overnight at 4℃. Wells of the plate were washed, blocked with 150 µl 1% BSA/PBST blocking buffer, and incubated with serial diluted recombinant TNF-β protein starting from 2000 pg/ml to 30.5 pg/ml for 1 hour at 37℃. The plate was washed and incubated with 50 µl per well of detect antibody HA721243 [PS01-19] (Biotin, 500 ng/ml) for 1 hour at 37℃. Then the plate was washed and incubated with 50 µl per well of Streptavidin-HRP for 0.5 hour at 37℃. Detection was performed using an Ultra TMB Substrate for 10 minutes at room temperature in the dark. The reaction was stopped with sulfuric acid and absorbances were read on a spectrophotometer at 450 nm. Capture/detect antibodies, recombinant TNF-β protein, Streptavidin-HRP are diluted in 1% BSA/PBST.
