Western blot analysis of Human IL-6 on recombinant protein with Mouse anti-Human IL-6 antibody (HA601050) at 1/500 dilution. Lysates/proteins at 50 ng/Lane. Exposure time: 30 seconds; 12% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (HA601050) at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Mouse IgG - HRP Secondary Antibody (HA1006) at 1:100,000 dilution was used for 1 hour at room temperature.
Human IL-6 Antibody (HA601050) in indirect ELISA. Indirect ELISA analysis of Human IL-6 was performed by coating wells of a 96-well plate with 50 µl per well of Human IL-6 antigen diluted in carbonate/bicarbonate buffer, at a concentration of 1 µg/mL overnight at 4℃. Wells of the plate were washed, blocked with StartingBlock blocking buffer, and incubated with 50 µl per well of a mouse Human IL-6 monoclonal antibody starting at a concentration of 20 µg/mL and serially diluting it to a concentration of 1.28 ng/mL for 2 hours at room temperature. The plate was washed and incubated with 50 µl per well of an HRP-conjugated goat anti-mouse IgG secondary antibody at a dilution of 1:10,000 for one hour at room temperature. Detection was performed using an Ultra TMB Substrate for 5 minutes at room temperature in the dark. The reaction was stopped with sulfuric acid and absorbances were read on a spectrophotometer at 450 nm.

Dilute the Capture Antibody (HA721167) to the working concentration 5μg/ml in PBS without carrier protein. Immediately coat a 96-well microplate with 100 μL per well of the diluted Capture Antibody. Seal the plate and incubate overnight at 4℃. Aspirate each well and wash with Wash Buffer, repeating the process two times for a total of three washes. Add human IL6 protein (HA210797) with 1:1 serially diluted starting from a concentration of 5,000pg/ml and incubate 45 minutes at room temperature. Add 100 μL of the HRP-conjugated Detection Antibody (PS00-15) at a dilution of 0.2μg/ml to each well. Cover the plate and incubate for 30 minutes at room temperature. Add 50 μL of Stop Solution to each well. Determine the optical density of each well immediately, using a microplate reader set to 450 nm.

The primary antibody (R1412-2, 1/5,000) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1/50,000 dilution was used for 1 hour at room temperature.

Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature and blocked with 1% Blocker BSA for 15 minutes at room temperature. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes.The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (R1412-2, 1/100) for 30 minutes at room temperature.
