Western blot analysis of Growth Hormone on human placenta tissue lysates. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST in PBS for 1 hour at room temperature. The primary antibody (ET1703-47, 1/500) was used in 5% NFDM/TBST at room temperature for 2 hours. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Then the plate was washed and incubated with 100 µl per well of SA-HRP for 0.5 hour at 30℃ with shaking. Detection was performed using an Ultra TMB Substrate for 10 minutes at room temperature in the dark. The reaction was stopped with sulfuric acid and absorbances were read on a spectrophotometer at 450 nm.
