Western blot analysis of IL-1 beta on different lysates with Mouse anti-IL-1 beta antibody (HA601002) at 1/1,000 dilution. Lane 1: THP-1 cell lysate Lane 2: THP-1 treated with 80nM TPA overnight then replaced with 100ng/mL LPS for 6 hours add 300ng/mL BFA for last 3 hours cell lysate Lysates/proteins at 20 µg/Lane. Predicted band size: 31 kDa Observed band size: 31/28/20 kDa Exposure time: 5 seconds; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (HA601002) at 1/1,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Immunohistochemical analysis of paraffin-embedded human tonsil tissue with Mouse anti-IL-1 beta antibody (HA601036) at 1/100 dilution. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HA601036) at 1/100 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

The concentrations of IL-1β were interpolated from the IL-1β standard curve and corrected for sample dilution. The mean IL-1β concentration were determined to be 27pg/mL and 1,615 pg/mL in unstimuated and LPS stimulated Raw264.7 cell extract.
