Background :

The two major cytoskeletal proteins implicated in cell motility are actin and myosin. Actin and myosin are constituents of many cell types and are involved in a myriad of cellular processes including locomotion, secretion, cytoplasmic streaming, phagocytosis and cytokinesis. Although actin is one of the most conserved eukaryotic proteins, it is expressed in mammals and birds as at least six isoforms characterized by electrophoresis and amino acid sequence analysis. Four of them represent the differentiation markers of muscle tissues and two are found in practically all cells.

Immunogen :

Synthetic peptide: KQEYDESGPSC conjugated to KLH, corresponding to amino acids 359-368 of Human beta Actin.

Specificity :

This antibody recognises beta and gamma actin in human samples. It probably also recognises all the other known forms of human actin. There is only one residue difference between the immunogen sequence (KQEYDESGPS) and the equivalent sequence in the other known forms (KQEYDEAGPS).This antibody detects a single clean band in Human, Mouse, Rat, Chicken and Drosophila samples. In Xenopus laevis a secondary band is detected at about 30kD. We are unsure whether this is cross-reaction with another actin isoform or merely non-specific. In Cow a doublet is detected, which probably represents different forms of actin.
