Transforming growth factor beta-1 (TGF-beta-1) is a stable polypeptide growth factor with diverse biological functions. Generally, TGF-beta-1 stimulates cells of mesenchymal origin and inhibits cells of epithelial or neuroectodermal origin. In normal tissue, TGF-beta-1 is involved in wound healing, angiogenesis, hematopoiesis, mammary gland development, bone metabolism and skin formation. It is also involved in many pathological findings, such as inflammatory, fibrotic diseases and tumor development. TGF-beta-1 stimulates cells by binding with TGF receptors type I and type II. The binding allows the phosphorylation of the receptors and the signal is propagated through the phosphorylation of Smad protein. Shown below is the latent form of TGF-beta-1 that is secreted by most cells. Pro-TGF-beta-1 consists of non-covalently bound dimeric LAP and mature TGF-beta-1, and it is linked to LTBP by a disulfide bond through Cys 33. The mature recombinant human TGF- 1 is cleaved from pro-TGFB1. It contains two homodimeric 112 amino acid residue polypeptides linked by a disulfide bond.
