Background :
Autophagy is a highly conserved bulk protein degradation pathway responsible for the turnover of long-lived proteins, disposal of damaged organelles, and clearance of aggregate-prone proteins. Autophagy is involved in various physiological or pathological processes, such as development, host defense response, cancer and neuronal degenerative diseases. It involves concerted action of more than 20 specific autophagy (ATG) proteins that mediate the formation of a double-membrane vesicle, the autophagosome, which engulfs the substrate and delivers it to the lysosome for degradation. Nbr1 (neighbor of BRCA1 gene 1) has been identified, together with p62, as a selective autophagy receptor, capable of simultaneously binding both ubiquitin and autophagy-specific LC3/GABARAP proteins. Nbr1 is recruited to ubiquitin-positive protein aggregates and degraded by autophagy through its interaction with the LC3/GABARAP component of the autophagosomal membrane. Such receptor proteins provide a molecular link between ubiquitinylation and autophagy, facilitating the selective targeting of various types of cargo, ranging from protein aggregates to membrane-bound organelles and microbes for autophagic degradation.
