Background :

The existence of a pancreas-derived hyperglycaemic factor has been known for over 70 years, but it was not until 1983 that the primary structure of the human glucagon precursor molecule was described. The human prepro-glucagon gene encodes a 180 amino acid, 21kDa protein which includes one copy each of the biologically active molecules glucagon and glucagon-like peptides 1 and 2 (GLP-1 and GLP-2). Pro-glucagon is cleaved in the pancreatic A-cells to release glucagon and the other pro-glucagon-derived peptides, but in enteroglucagon (EG)-containing L-cells of mammalian gut the enzymatic conversion of ‘glicentin’ to ‘pancreatic’ glucagon does not occur. Recent work has shown that prohormone convertase-1 (PC1) is capable of processing pro-glucagon to produce the intestinal peptides, glicentin, oxyntomodulin and GLPs, whereas PC2 processes pro-glucagon to glicentin but does not produce glucagon either alone or in combination with other enzymes or with other known molecular chaperones.

Uses :

Test tissues: Mammalian endocrine pancreas (A cells) and colon (EG cells) are heavily labelled with this antiserum. Fixatives: All standard cross-linking agents may be used. Processing: Paraffin wax- and resin-embedded tissue has been used successfully. Dilution range: 1:1,000-1:8,000, using overnight incubation and indirect immunoperoxidase procedure (such as PAP7 or ABC-peroxidase). 1:200-1:1,200, using overnight incubation and indirect immunofluorescence procedures. Specificity: Immunostaining may be abolished by pre-incubation with 10nmol synthetic or natural glucagon per mL diluted antibody. Glucagon-like peptides 1 and 2 at the same concentration have no effect on immunostaining.
