Background :
Eukaryotic elongation factor 2 (eEF2) mediates the GTP-dependent translocation of nascent peptides from the A-site to the P-site of the ribosome relative to mRNA after addition of each amino acid to the nascent chain. Phosphorylation of eEF2 within the GTPbinding domain at Thr56 by eEF2 kinase inactivates eEF2 and results in the termination of mRNA translation. Conversely, eEF2-regulated protein synthesis can be stimulated by dephosphorylation of eEF2 at Thr56 by protein phosphatases such as PP2A, and/or by upstream kinase inactivation via the mTOR pathway.
