PLCγ-1 (Phosphoinositide-specific phospholipase C) is a 135 kDa isoenzyme belonging to the PLC family of proteins that hydrolyzes phosphatidylinositol 4,5-biphosphate to produce the second messengers IP3 and diacylglycerol (DAG). PLCγ-1 is ubiquitously expressed and modified by phosphorylation at two key tyrosine residues, Tyr783 and Tyr771, in response to non-receptor and receptor tyrosine kinase stimulation. Tyr783 phosphorylation is required for PLCγ-1 activation, and regulates EGF-induced association of PLCγ-1 with Akt.
