Background :
AIB1 is a member of the p160/steroid receptor coactivator (SRC) family, named for its common amplification in breast cancer. Members of this family include a defined nuclear receptor box motif (LxxLL) that is critical for binding to nuclear hormone receptors, and two C-terminal activation domains that bind secondary coactivators such as the p300/CREB binding protein (CBP) and CARM-1 to confer acetyltransferase or methyltransferase downstream signals, respectively. AIB1 binds to estrogen receptor (ER), and its overexpression in hormone-responsive cancers and frequent amplification in breast and other cancers make it a focus for the treatment of cancer and the study of hormone receptor signaling.
