Background :
TOR1 is one of three genes (FPR1, TOR1, and TOR2) mutations of which confer rapamycin resistance in the yeast Saccharomyces cerevisiae. The proteins encoded by the TOR1 and TOR2 genes are large (250 kDa) in size, have homology to the phosphatidylinositol and protein kinases, and TOR2 possesses PI 4-kinase activity. The TOR proteins and their mammalian homolog (FRAP, mTOR) form a complex with a 12 kDa cytosolic protein, FKBP-12 and rapamycin to effect arrest in the G1 phase of the cell cycle. The G1 block is mediated by the activation of ribosomal p70S6K and cyclin dependent kinases. The cell cycle arrest activity requires the presence of the intact kinase domain in the TOR protein and in particular a conserved Ser residue within the domain. The human hTOR (mammalian or mTOR) protein shows 39-43% homology with the S. cerevisiae proteins TOR1 and TOR2. Ser2448 has been identified as a PKB-mediated phosphorylation site.
