Background :

Keratocan is an extracellular matrix protein that belongs to the small leucine-rich proteoglycan (SLRP) family that also includes lumican, biglycan, decorin, mimecan, and fibromodulin. Members of this family are known to regulate cellular processes, such as proliferation and modulation of osteoprogenitor lineage differentiation. Keratocan is differentially expressed by osteoblasts and shows little or no expression in osteocytes. Lumican and Keratocan are the major keratan sulfate (KS) proteoglycans in corneal stroma. Both proteins are critical for normal cornea morphogenesis during embryonic development and maintenance of corneal topography in adults. It is shown that homozygous Keratocan knockout mice display abnormal corneas, significantly decreased rates of bone formation and mineral apposition, larger fibril diameters, and disorganized fibril spacing. Keratocan mutation leads to cornea plana in humans, which is often associated with glaucoma. Experimental evidence indicates that lumican may have additional biological functions, such as modulation of cell migration and epithelium-mesenchyme transition in wound healing and tumorgenesis, aside from regulating collagen fibrillogenesis.
