Background :

Protein disulfide-isomerase, also known as PDI, is an abundant component of the ER that catalyzes disulfide bond formation in the biosynthesis of various secretory and cell surface proteins. It is a multifunctional polypeptide identical to the β-subunit of prolyl 4-hydroxylase, a α2β2 tetramer that catalyzes the formation of 4-hydroxyproline in collagen by the hydroxylation of proline residues in peptide linkages. PDI has two thioredoxin-like regions, each containing a Cys-Gly-His-Cys sequence that may represent two independently acting catalytic sites of PDI activity. The C-terminus of PDI has a KDEL sequence, which acts as retention signal for proteins of the ER lumen. The PDI/β-subunit of prolyl-4-hydroxylase is required for its retention in the ER.
