Background :

Glycogen synthase-3 alpha (GSK-3 alpha) is a ubiquitous constitutively-active multifunctional kinase. GSK-3 alpha phosphorylates serine and threonine residues on its target substrates, most of which are “primed” or prephosphorylated. It is most widely researched for its interaction with glycogen synthase which is deactivated after phosphorylation by GSK-3 alpha; this activity blocks the synthesis of glycogen and plays an important role in the regulation of glycogen. GSK-3 alpha is also involved in the Wnt/β-catenin, Notch, and Hedgehog signaling pathways and affects a wide range of cellular processes including cell-cycle regulation, transcription, and apoptosis. GSK-3 alpha also phosphorylates tau proteins and may contribute to the generation of APP-derived amyloid plaques which are involved in the pathogenesis of Alzheimer’s disease. GSK-3 alpha is also investigated for its role in type 2 diabetes, and cancer.
