Background :

erbB-3/HER-3 is a receptor tyrosine kinase (RTK) and is a member of the EGFR/ErbB family of receptor tyrosine kinases. Four subfamilies make up the ErbB family of receptors: erbB-1 (EGFR), erbB-2 (HER2/c-neu), erbB-3 (HER3), and erbB-4 (HER4). erbB-2 is membrane bound and able to bind its ligand, but it does not have an active kinase domain to transmit this signal. In order to transmit the signal, erbB-3 must first form a heterodimer with other ErbB family memebers. This heterodimerization then leads to proliferation and differentiation. erbB-2 also binds Phosphoinositide 3 Kinase (PI3K) through six tyrosine phosphorylation sites that PI3K can bind resulting in the activation of AKT/PKB. erbB-3 has been shown to be amplified in numerous types of cancer, including breast and prostate.
